Quantification of the interaction of lysolecithin with phosphatidylcholine vesicles using bovine serum albumin: relevance to the activation of phospholipase A2.

The activity of soluble phospholipase A2 to hydrolyze phosphatidylcholine vesicles increases abruptly after a lag time of several minutes. The onset of this apparent activation event probably results from the accumulation of a threshold mole fraction of the hydrolysis products (lysolecithin and fatty acid) in the bilayer. One important observation relevant to the mechanism of this activation process is the biphasic dependence of the lag time on vesicle concentration. To test whether this dependence can be attributed entirely to the strength of partitioning of the lysolecithin into the phosphatidylcholine bilayer, we estimated the apparent partition coefficient of lysophospholipid in the membrane of phosphatidylcholine vesicles. Based on competition between bovine serum albumin and the vesicles for the lysophospholipid, we estimated the partition coefficient to be about 5.10(-7) for palmitoyl lipids at 39 degrees C and about 9.10(-7) for myristoyl lipids at 22 degrees C. These values were able to rationalize the behavior of the lag time with dipalmitoylphosphatidylcholine vesicles, but they were unable to predict the behavior with dimyristoylphosphatidylcholine. Therefore, it appears that the complete dependence of the lag phase on vesicle concentration must be explained by additional means such as the possible contribution of nascent fatty acid or previously proposed kinetic activation mechanisms.

[Comparative characteristics of changes in the fatty acid spectrum of high density lipoproteins in male patients of various age with appendicitis]

The fatty acid spectrum, the content of cholesterol of total high-density lipoproteins (HDLP) and high-density lipoproteins3 as well as the activity of lecithin-cholesteryl-acyl-transferase (LCAT) were investigated in mature-age males (periods I and II) with appendicitis. The intensity of endogenous production of polyunsaturated fatty acids was concluded to be reducing in mature-age (periods I and II) males. As for the mature-age males of period I, the consumption of polyunsaturated fatty acids was found to be increasing and the production of endogenous cholesterol was found to be decreasing in them. The transformation of HDLP3 into HDLP2 was found to be declerating in the mature-males of period II, while the cholesterol-acceptor ability of HDLP3 was going up in the males of latter category.

Growth of rat mammary tumor line 64-24 in liposome-supplemented defined medium. I. Effect of liposome B components on colony growth.

An improved serum-free medium has been developed that supports growth of rat mammary tumor line 64-24 with far less protein supplementation and with a much smaller inoculum than previously possible. An initial survey showed that MCDB 202 supported clonal growth with 1% dialyzed serum. The remaining serum was then replaced with 5 micrograms/ml insulin, 10 ng/ml epidermal growth factor (EGF), 1 micrograms/ml hydrocortisone, 50 ng/ml ovine prolactin, and 5 micrograms/ml liposome B (a mixture of soy lecithin, sphingomyelin, cholesterol, vitamin E, and vitamin E acetate in liposome form). Insulin and EGF are required and growth is improved by hydrocortisone and prolactin. Estradiol is stimulatory in the absence of liposome B. With adequate iron supplementation, transferrin has no effect. Liposome B increases growth rate substantially. Most of the growth stimulation can be replaced with phosphatidylethanolamine or sphingomyelin.
